I think the all the runes (except for Hel of course) in this new Splash image, are NEW runes to the Diablo Series and can be expected to be used in the new game.
They may be similar to other games, because well, duh, there made by same company.

The creature we see is organic, it is not a helm, or a robot. I looks protosish, but that may be a way to confuse us some more.
My guess would be either one of the lesser evils (Azmodan, Kabraxis, etc..) escaping.

I think the hugeness of these threads makes the acquisition of actual information quite difficult and annoying.
At the same time, the mere fact that there is this much fanaticism has to be a good sign to Blizzard that the fan base is QUITE alive and kicking.
