Both D1 & 2 had some pretty incredible quest, who could forget the butcher, king leoric, or breaking the seige of harrogath ?
Each was unique, and each was incredibly fun to play.
Blizzard has some pretty high standards to break if they want to outdo the quests from their past games, what do you think they'll come up with?
Furthermore, what do you specifically want to see in a quest ? What will it involve doing ? Will it be a rescue mission, a "kill this guy" kind of mission, or a massive over-arching quest like making Khalim's Will ? Make your description of the quest as detailed as you want !
Personally, I want to see a quest where you have to venture through a town under seige by Fallen, having to fight your way through streets and rubble to find survivors, kill enemies, and eventually kill the leader of the seige. Much like the seige of harrogath quest, but in a more urban setting.
Anywho, what are your ideas ?

One mother witnessed the beast dragging her daughter off in a certain direction and tells you it was heading toward a old well outside of town. You light a second torch and toss it in front of you a few feet. The glow reveals a horrific site. Mounds of bones, now obviously childrens bones some still wearing partially torn clothing and some still covered with rotting bits of meat.
Further ahead you hear a sobbing and then a scream. You walk toward the sound, all is silent now except the sickening sound of raw meat being feasted upon. The beast standing over the small child's body turns and looks at you it's eyes burning a shade of crimson. It's standing just at the edge of the light but that's enough for you to see the nearly six foot tall figure. The blood covering it's mouth, the large dagger like teeth and the claws at the end of the demons hands that could easily be small swords. It makes a low guttural growling sound at you and begins to charge you, it's skin a sickly dark green, and then the beast is upon you and the battle is at hand.
I'd love something as gruesome as that.
That right there is an example on how to do things right, if everybody posted an idea like this, this thread could go on for months ^^.
Anyways, get as in depth as you want, and don't feel afraid to get gruesome !

I like the idea of fighting for hell though, while I doubt we would have an act or a series of quests devoted to it, we may have to kill an angel here and there since apparently heaven is going to play a bigger part in D3.

I like the idea of a quest inside of a dungeon where you each room you previously were in floods and you cant go back. The flloding is activated by traps and you could accidently fllood the room your in if you are too slow. Maybe place this in the last level of the game along with my idea of the shifting walls.

The shifting walls idea was beyond awesome... like I'm really going to be sad if we don't get that In D3, and I'm going to direct all the blame at you ^^.
The flooding rooms sounds pretty awesome though, would create a really suspenseful feel to the quest.
Maybe you would be battling a boss, and if you didn't weaken him enough in each room, you drown ?
Oooor....
Maybe you would have to like... save books from becoming soggy ?

Well, since Habacalva could do it, I want to visit the Elemental Planes. Not entirely sure what that would imply, but to travel the very birthplaces of magic, the domains of the Elementals like the Hydras or Gloams. I've always loved my casters, and, to me, this would be akin to the Barbarian traveling the heights of Mount Arreat.
-I would like a quest which starts in ACT1 and continues on till the game is finished.
-I want the outcome of this quest to depend on the choices you made along the way:
so 'you reap what you sow'
-I want at least 20-30 variations of rewards so it is even worth playing the game over just
to get a different ending.
-I want this quest to have an amazing story which explains some parts of the main story
in more detail.
That is my dream Diablo quest.

That quest idea sounds pretty badass, it would add so much variation to the game, may be difficult to mesh in with multiplayer though. Perhaps when you join games, you join the world that the player who started the game created? Like if they chose to kill deckard cain (random example here), and you chose to let him live, then in the game you would have to play with the choice that the other player made ?
It would be pretty hard to make this work in multiplayer if it was a main quest, because everybody would have a different version of what happened to their character. But if it was a massive side quest, it would work perfectly ^^.
I hope we get something like this, would make D3 so much more replayable :D.

I would like to see a quest where there is a big town or city under siege, and the demons have a special very powerful weapon that they want to unleash on the following day, and your mission is to go into their camp at night, find the weapon and destroy it. Of course you could do this without fighting anyone but the weapon's guard, or you could wake up as many monsters as you want either on purpose or by accident. Also, if you don't so it during the night, the army attacks in the morning and you would either have to kill every monster, or watch the city get destroyed along with insignificant NPCs that aren't part of the main story, but would be useful to you if they were alive.
Man, an infiltration quest like that would kick ass ^^.
You would probably have to get some kind of massive reward for infiltrating though, as most players would just slaughter their way through the whole camp.
Maybe Azmodan could appear in a burst of smoke and be all "Huzzah ! You spared my minions, here, have my microwave oven."

In all seriousness though, I think this is a great idea.
Would add a nice alternative quest, instead of the usual "You HAVE to kill this guy" type of quests we get in the diablo series. Not to say those types are bad though, I still love them to death, but some quest variety is always a good thing.

i would love to do some quests under the water. i think sunken treasure hunting would be cool whattya think about that?
Haha, that would be pretty sweet ^^.
Could have a massive network of caves with flooded sections, dry parts, and completely submerged tunnels, and you'd have to swim through the submerged sections fast enough of drown ^^.
Would be pretty neat to see what they came up with for fish in these areas, we could end up seeing something along the lines of the slaughterfish in the elder scrolls series.

My ideal quest was already mentioned and that would be to play as an evil character. I would love to go in as Dark Cultist or Fallen Shaman and lay siege to a small town with a horde of demons and undead. Would be cool if it was the first quest you do even before creating your character and it could help explain the events of the first act.

I really like that idea !
Having that as a first quest would be quite interesting, might get annoying after some time though, so a skip function would be needed.
Would add some backstory to the game, and open up some interesting gameplay options, like being able to trasnform into one of those demon/cultist things.

Well technically you don't fight for heaven in Diablo. You fight for the human world. Sanctuary.
But yeah some quests where you actively have to fight a angel is always welcome to me.
